بررسی آزمایشگاهی آثار آنتی اکسیدانی، آنتی رادیکالی و آنتی دیابتیک و ممانعت از تشکیل محصولات نهایی حاصل گلیکیشن پیشرفته توسط دارچین

چکیده

امروزه گیاهان دارویی به خاطر نقشی که در پیشگیری و درمان بسیاری از بیماری‌ها دارند، مورد توجه قرار گرفته‌اند. هدف از این مطالعه، ارزیابی آزمایشگاهی خواص آنتی-اکسیدان و ضد دیابتی‌دیابتیک گیاه دارچین می‌باشد.
عصاره‌ی آبی دارچین با روش خیساندن تهیه شد. غلظت‌های 25/0، 5/0، 1 و 2 میلی گرم در میلی لیتر از عصاره فراهم گشت. خواص فیتوشیمیایی دارچین با اندازه-گیری فنل، فلاوونوئید، ظرفیت تام آنتی اکسیدان، قدرت پاک کنندگی رادیکال، توانایی کلاته کردن فلز و اندازه گیری گروه‌های تیول معین شد. فعالیت آنتی‌دیابتیک نیز توسط توانایی مهار تشکیل فروکتوز آمین و محصولات پیشرفته حاصل از قندی شدن به ترتیب توسط اسپکتروفوتومتری و اسپکتروفلورومتری سنجیده شد. توانایی عصاره در فراگمانتاسیون آلبومین گلیکه با SDS-PAGE سنجیده شد. مقدار فنل و فلاوونوئید توسط روش‌های استاندارد اندازه گرفته شد. داده ها با ANNOVA یکطرفه و با نرم افزار SPSS آنالیز شد.
مقدار فنل دارچین mg GAE/g83/1± 34/31 و مقدار فلاوونوئید آنmg QE/g 37/1 ±89/19 معین شد. در همه روش‌ها، فعالیت‌های آنتی اکسیدان و آنتی رادیکالی دارچین ، به صورت وابسته به غلظت تایید شد. این گیاه دارای خاصیت آنتی دیابتیک است و تشکیل فروکتوزآمین و محصولات پیشرفته حاصل از گلایکیشن را کاهش می‌دهد و همچنین فراگمانتاسیون آلبومین گلیکه را مهار می‌کند.

چکیده [English]

Nowadays medicinal plants are in the focus of attention because of their roles in prevention and treatment of many diseases. The aim of this study was in vitro assessment of antioxidant and antidiabetic properties of Cinnamon zeylanicum.
Aqous extract of cinnamon were prepared by maceration method. The 0.25, 0.5, 1 and 2 mg/ml concentration of this extract were prepared. The phytochemical properties of Cinnamon detected by measuring the phenol and flavonoid, total antioxidant capacity, radical scavenging power, metal chelating ability and thiol groups assay. Antidiabetic activities also were assayed with fructosamine formation inhibition and advanced glycation end products(AGEs) inhibition abilities by spectrophotometric and spectroflurometric methods respectively. Ability of the extracts in preventation of glycated albumin fragmentation was assayed by SDS-PAGE. The content of phenol and flavonoid were determined by standard methods. The data were analyzed by one way ANOVA procedure according to SPSS software program.
Cinnamun exhibited 31.34±1.83 mg GAE/g extract of phenol contents and also 19.89±1.37 mg QE/g extracts of flavonoid content respectively. All methods confirmed the antioxidant and antiradical effects of cinnamon in a concentration related manner. This plant has antidiabetic properties and reduced fructosamine and AGEs formation and also could inhibit glycated albumin fragmentation.
Cinnamon has antioxidant and antidiabetic properties and could prevent complications of diabetes mellitus. According to these properties, we can suggest using this plant in diabetic patients.
